Bacteria are microorganisms that grow everywhere. Are you interested in the growth of bacteria? We can collect them and culture them in petri dishes to observe. So, how to grow bacteria in a petri dish? Read this blog to learn about it.
Part1.Pre Lab Preparation for Growing Bacteria Experiment
Part2.Operating Steps for Culturing Bacteria
1. Pour 10g of agar into 330ml of water and stir the solution until it is completely dissolved.
2. Heat the container containing the solution in the microwave for 1 minute or adjust it according to your microwave power. Please ensure that the agar powder is completely dissolved and the liquid color is clear. Note: please do not let the agar solution boil when heating.
3. Cool the solution for a few minutes and pour it into Petri dish. Let the solution form a layer at the bottom of Petri dish and cover the lid.
4. Leave Petri dish for 30 minutes to 2 hours until the agar solution cools and hardens.
5. Collect samples with cotton swabs. You can collect bacteria from any surface.
6. Draw a squiggly line in the agar with a cotton swab dipped in the sample. Note: Don't scratch the agar layer
7. Store the Petri dish in a warm, dark place. The ideal temperature for bacterial growth is 70-98 degrees Fahrenheit (20-37 degrees Celsius). When placed, Petri dish can be stored upside down, so that the growth of bacteria will not be disturbed by any water droplets.
