Glass slide:

The first element to get the highest quality slides is to choose the best microscope slide. Some of the glasses in the microscope slide contain impurities. These pollutants floating in the glass may produce false color or fluorescence, which will reduce the contrast and clarity.

The microscope slides must be made of optical quality glass. Some of the best microscope slides are made of soda-lime glass or borosilicate glass. Sometimes transparent plastics of special optical quality are used.

Dry mounting is the simplest mounting method. It just puts the thin, light transmission subject on the slide. Usually, a cover slide is placed on the top to keep it flat and keep the objective of the microscope from contaminating the subject.

A wet or temporary mount means that the subject is immersed in water or another liquid on a slide. Wet mounts are usually used for liquids related to the nature of the subject (e.g., watching swimming protozoa). Cover slide is almost always used to flatten the object and avoid the refraction effect of the liquid.

Mounting media:

The mounting medium can be water. Sometimes glycerol (slightly denser than water) is used.

The permanent mounting usually adopts specially designed materials to keep the thickness and fixation of the specimen for a long time, to reduce corrosion and pollution.

The mounting medium shall not act as a lens as this will increase additional light refraction and distortion. The installation medium must be selected so that its refractive index is the same as that of the sample. If the two refractive indices are too different, the points where the medium strikes the sample will be shown as dark areas, which can hide important details.

Stain:

Stains will be combined with special features of cell anatomy to make them more visible under the microscope. There are many kinds of stains with different uses. The quality of your slides usually depends on the proper use of stains.
